A. ProblemThe purpose of this lab is to observe how enzymes act on substrates to produce products. We will observe how enzyme activity is altered when the enzymes environment's pH (also known as potential hydrogen) is changed.
B. HypothesisI believe that if i were to make lactase reaction more acidic that the products formed will preform much less. I believe this because based of my prior knowledge of learning about this; enzymes like to be in a certain environment so that it could do its job. So with knowing this if I had acid in it the acid may contradict with the base which may lead to a slower performance rate,
I believe that if I were to make the pH of lactase more basic it will preform a lot better. I believe this because also based of my prior knowledge about learning about this; enzymes like to be in a certain environment so that it could do its job. Therefore if its in a basic solution (which milk is basic). It would be in an environment in which it is comfortable in and will preform a lot more.

F. Conclusion
- As you add more of the substrate, more products would be formed. This when graphed will be linear.
- The maximum initial reaction rate for lactase at the pH of 7 350(10^6) molecules formed per minute. This was in trial four.
- The reason why the maximum of the initial reaction rate cannot be reached at the low lactose concentration is because there is a less amount of substrate.
- My data displays that lactase like to be at the pH level of 3
What I expect to occur is that the lactase will work better in a base solution rather than an acid solution. This is because enzymes like to be in an environment in which they're comfortable with. Therefore if that lactase (which is a base) is in an acidic solution I believe that it will be spending to much time either fighting it or adapting to it that it may not preform well. If it were in a base solution I certainly believe that it will preform much better. The reason being is because it is in a solution in which it is comfortable with. I believe that since it will be in a environment that its comfortable with that it won't be spending time either trying to fight it or adapt. It will go right along with it and preform well.
My Lab Report about
Hypothesis: I have conducted a hypothesis that involves lactase enzymes, and the pH measurement. I believe that when lactase is in an acidic solution it will not preform well. I believe this because based of of my prior knowledge lactase is a basic solution. I also remember that lactase preforms well in an environment that it is comfortable with. Therefore a basic enzyme like lactase in my opinion will not preform best at acidic pH. I believe that when its in an enviorment that it is comfortable with which is basic it will preform at its greatest.
Materials:
Procedure:
Results: Make sure that you are keeping watch of this as this takes place. If you notice some changes that occur make sure that you write it down. It would be highly recommended that as you write down these observations that you create a graph. Preferably a line graph that would be the best kind of graph to make.
Conclusion: Therefore with my hypothesis that involved lactase enzymes, and the pH measurement. Off of my observations my hypothesis was partially correct. It was correct in that acids would not be the best type of enviornment for lactase to be in. However I was incorrect and lactase preformed better at a neutral state. What I could change to improve this lab next time is to try to conduct this in different temperatures and see if it will have any effect. I learned that enzymes preform better at a neutral state. This was evident based on the graph
Materials:
- I used 5 test tubes.
- A total of 77.5 grams of lactose all together.
- I used 5 tabs of the lactase enzyme.
Procedure:
- Make sure you have a journal and a pencil or pen
- Second you must obtain 5 test tube for this is where the experiment will be conducted.
- Third you must vary the pH of each of the test tubes. Have test tube 1 at the pH of 3, Test 2 at the pH of 5, have test tube 3 at the pH of 7, have test tube 4 at the pH of 9, have test tube 5 at the pH of 11. Do not change this at any time
- It is vital that you add the lactase enzyme to each of the 5 test tubes.
- You will be conducting 5 trials. For trial one add only 0.5 grams of lactose, for trial two add only 1.0 gram of lactose, for trial three add only 2.0 grams of lactose, for trial four only add 4.0 grams of lactose, and for trial 5 only add 8.0 grams of lactose.
Results: Make sure that you are keeping watch of this as this takes place. If you notice some changes that occur make sure that you write it down. It would be highly recommended that as you write down these observations that you create a graph. Preferably a line graph that would be the best kind of graph to make.
